I spent a lot of time in Winston-Salem on business and my recommendations:
Lodging: Stay in the Hanes Mall area.
Food: For a nice casual restaurant, The Village Tavern.
If you want some really good BBQ you have to try Camel City BBQ downtown.
The Brisket & Smoked Chicken Wings are great. Kind of weird, they have their own parking lot but you need to enter your license plate number inside at a little kiosk or you'll get a bill for $25 in the mail in a few weeks.
